The problem is that your scanner is not based on the WIA API to interact with Windows & other programs (but on the TWAIN one), and Paint.NET only implement WIA to acquire from a scanner, so you just can't acquire on PdN. You can delete the layer you've selected.

I see this in some photoshop tutorials too. It's not included in Paint.NET and i don't know if it will be someday.
