Is there any way I can do this in Paint.NET or is there some kind of alternative that would allow me to do something similar? Thanks. What I usually do is add a new layer and use the Grid plugin to render a grid pattern at the correct size and use that as the guide. I always found it to be one of the most useful in Photoshop. I guess it's about time I downloaded the source for PDN and actually wrote the plugin myself. There needs to be more time in the day. The main reason this hasn't been implemented yet is that guides are of limited use without snap-to functionality. Currently this would require every tool to implement the snapping functionality, which ends up being a fragile and taxing way to do things. I would much prefer to have this implemented at, say, the input layer, so that it is automatically handled by all tools. It just hasn't been done yet.
