There is no pressure sensitivity feature in paint.net as of now, so (also as Jim said) the result will look just like you used a normal mouse. However, if you set the color opacity to something low (ex: 20) you can use the speed of the brush movement as "negative pressure." The faster you draw, the lighter the color. It works much better than the mouse because the pen allows for better precision.
