Selecting in a png file

I always wondered if there was an easier way of selecting an object in a png file. Now I just use the Magic wand but I wondered if there was a way where you could do it in one time. 

I'm not talking about an extra layer because I know that trick but I just only need the selected part and not the object itself.

Without layers you need to separate the target element from the background. In most cases the magic wand is the most appropriate tool. The exception is where the element is a regular circle/ellipse, square/rectangle or a reasonably simple combination of these shapes.
