No I have a scale on the image and the measurement in Paint.NET of that scale is not the same as the scale Is there a possibility to calibrate the measurement tool in Paint.NET?

I measure a distance on an image on which I have scale (medical microscopic slide)

is anybody knows why while measuritrng a known distance, I do not get the correct lentgh in centimeter?

Is there a way to calculate length and area in milimeters by tracing the borders of a shape? How one overcome the scaling problem?
