Access is denied

Looks like you opened an image file that is on a CD, and then tried to save the modified image back to the CD.

Or if not a CD, it was a file that was otherwise set to read-only.

See that padlock icon on your file there in your screenshot? That means you are not the owner of the file, or otherwise don't have write permissions.

 

Click on the "Security" tab, and take a look at the details it gives you.

This is pretty self-explanatory. I assume you're just looking a confirmation?

In paint.net, click on File, and then click on Save As.

Did you install this from the Windows Store? If so, you won't be able to modify files within the "application package," which is what it looks like you're trying to do. This will not work. At all.
